Introduction of luminescent products and markers

When the energy generated by the chemical reaction of the substrate is released in the form of light, we call this process chemiluminescence. Luminol is one of the most commonly used chemiluminescent reagents. When oxidized by peroxide, it produces an excited state product, aminophthalate. This product decays to a low energy state while releasing photons. Patented additives increase the luminescence intensity and duration (stability) of the luminol reaction, making enhanced chemiluminescence (ECL) possible.

Chemiluminescence has become the preferred method of detection for most protein laboratories. These advantages are as follows: 1. The best imaging results can be obtained by multiple exposures using chemiluminescence; 2. The detection reagent can be completely removed, and other proteins of interest can be performed. Detect and image, or optimize the detection conditions; 3, has a wide linear response range, so that a wide range of concentrations of protein can be detected and quantified; 4, chemiluminescence has the highest sensitivity in all detection methods.

In the chemiluminescence analysis, if a suitable enhancer is further added, the sensitivity is improved and the luminescence time is prolonged, and the reproducibility of the measurement can be improved. The experiment proves that the phenolic derivative of the para-phenolic derivative has obvious luminescence enhancement effect, and its luminous efficiency can be increased by several tens of times, and the luminescence time is also effectively extended.

Acridine esters or acridine sulfonamides are used in chemiluminescence immunoassays (CLIA), and the commonly used system is the Acridinium ester/H ₂ O ₂ system. That is, the antibody or antigen is labeled with acridinium ester or acridinesulfonamide, and HNO ₃ + H ₂ O ₂ and NaOH are used as the luminescent start reagent. Some surfactants such as Triton X-100, CTAC, Tween-20 are added to the luminescent starter reagent to enhance luminescence.

Bioluminescent organisms are widely distributed in nature, including bacteria, fungi, fish, insects and the like. The various enzymes that catalyze the bioluminescent reaction in these bioluminescent organisms are called luciferases, and the substrate is called luciferin. D-luciferin is the most widely used substance for biological luminescence examination. It is the core component of fluorescein detection kit and is widely used in biology, medicine, food, health and environment. D-luciferin, D-luciferin potassium salt, D-luciferin sodium salt, as a substrate for Luciferase, are commonly used in the following experiments:

3. Determination of adenosine triphosphate (ATP)

4. Other luciferase and its gene related analysis and research work
