Solid phase extraction method for detection of β-agonist agonist in animal-derived foods (PolybaseTM MCX)
First, the purpose of the experiment
In this study, solid phase extraction was used as a pretreatment method for pork samples, and LC-MS/MS method was used as a detection method. The method can simplify the pretreatment process of pork samples, save the use of organic solvents, and is easy to operate.
Second, the experimental target
Five β-receptor agonist substances: clenbuterol (CAS: 37148-27-9), salbutamol (CAS: 18559-94-9), ractopamine (CAS: 97825-25-7), terbuta Lin (CAS: 23031-25-6), Tobbutro (CAS: 41570-61-0).
Third, the scope of application
The method is applicable to the detection and confirmation of β-agonist LC-MS/MS in animal-derived foods.

PolybaseTM MCX Solid Phase Extraction Column 6mL/200mg.
Sixth, experimental methods
1, sample extraction
Weigh 10.0 g of the homogeneous sample (accurate to 0.01 g), add 15 mL of acetic acid-ammonium acetate buffer solution of pH 5.2, homogenate for 1000 min at 1000 r/min, and then add β-glucuronidase/ 100 μL of the aromatic sulfonate solution was hydrolyzed overnight at 37 ° C ± 1 ° C. After taking out the cooling, it was centrifuged at 9500 r/min for 10 min at 10 °C. Take all the supernatant and purify it.
2, SPE column purification
(1) Activation: Activation was carried out by adding 6 mL of 5% methanolic ammonia, 6 mL of methanol, 6 mL of water, and 6 mL of 0.1 mmol/L perchloric acid (pH 4.0) solution.
(2) Loading and elution: When the solvent liquid level reaches the surface of the column adsorption layer, immediately pour the above-mentioned solution to be purified, pass the column, discard the filtrate, rinse with 2mL methanol, 2mL 2% methanol aqueous solution, and finally use 7mL5 % methanol ammonia eluted.
(3) Re-dissolution: The eluate was blown to near dryness under a slow nitrogen flow condition, dissolved to 1.0 mL with methanol-0.1% formic acid aqueous solution, and vortexed for 1 min for LC-MS/MS measurement.

1. 0.5 ppb pork matrix spiked recovery results: The recovery rates of the five analytes were all greater than 80%, and the RSD value was less than 5, meeting the standard requirements.
